A puncture is a small hole made by something sharp and pointed, like a nail or a pin. The most common way you'll hear it is about tires - if you run over something sharp and air leaks out, that's a puncture (American English often just says 'a flat tire'). Doctors also use it as a verb for making a small hole in the skin on purpose, like taking a blood sample. As a verb, to puncture something can also mean to ruin someone's confidence or a happy mood, the way a needle bursts a balloon.
