To ask for God's protection or favor over someone or something, often said in prayer or at a religious ceremony. In everyday talk, English speakers say 'Bless you!' right after someone sneezes - it's just a polite little habit, not really religious anymore. 'Blessed' (the adjective) means lucky or fortunate, as in 'I feel blessed to have such good friends.'
